Occupational Summary

Conduct a variety of investigations in the planning, development and implementation of experimental procedures within the Molecular Genomics Core lab at Duke University for the generation of spatial expression data. This position includes both the development and application of transcriptomics and histology techniques.

Work Performed

Collaborate with investigators from a variety of disciplines to aid in the planning, development and implementation of experimental procedures associated with spatial expression analysis. The position involves applying existing techniques and developing novel gene expression profiling approaches in the analysis of materials under experimentation. The position will require the applicant to meet and consult with customers to present core services and provide advice on their projects.

The applicant will be required to evaluate and interpret collected data and prepare reports and analyses setting forth progress, and appropriate recommendations for the improvement and conclusions associated with the data.

The applicant will need to maintain laboratory equipment and perform preventative maintenance as necessary, and maintain enough inventory of materials, supplies and equipment for performance of duties.

The applicant will be expected to review journals, abstracts and scientific literature to keep abreast of new developments in the field of spatial biology and to obtain information regarding previous experiments to aid in the design and development of original procedures and techniques.


Prepare human tissue specimens for the generation of existing and novel experimental approaches. These procedures include (but are not limited to) tissue processing, OCT embedding, paraffin embedding, freezing, DNA/RNA extraction, sectioning and staining. Tissue sections will be the material from which existing and developmental gene expression data in the spatial context will be generated.

The person in this position must have significant knowledge and experience to operate independently for the histology tasks.


The applicant will be trained in the running of single-cell RNA-Seq and spatial transcriptomic analyses. The spatial analyses will include running the existing 10x Genomics Visium platform as well as becoming intimately involved with the development of molecular level gene expression spatial profiling.

Required Qualifications at this Level


Work requires a bachelor's degree in botany, biology, zoology, psychology or other directly related scientific field.


Work requires two years of research experience. A related master's degree may offset required years of experience on a 1:1 basis, e.g., a two year master's degree in lieu of two years of experience.




  • Tissue processing - freezing, fixing, embedding in both OCT and FFPE

  • Sectioning both OCT and FFPE tissue blocks

  • Knowledge of the anatomy of a variety of tissues, and ability to work with the investigator to identify and isolate the region of interest

  • H&E and IF staining and imaging

Knowledgeable and proficient in basic laboratory techniques

Must be able to manage multiple and rapidly changing priorities and have ability to quickly learn new skills

Must be detail-oriented, well organized with strong communication skills and can work in an interactive team environment
