Position Description:

Exelixis is seeking a highly self-motivated, interactive, and productive hands-on PhD scientist to join the Pharmacology team working towards discovery of anti-cancer agents for the treatment of targeted indications. The successful candidate will have strong knowledge and extensive experience in the development of analytical methods for the characterization of proteins and peptides (intact mass and peptide mapping) using state-of-the-art LC-HRMS instruments, including Agilent RapidFire/QTOF and Sciex UHPLC/ZenoTOF systems.

Position Requirements:

ESSENTIAL DUTIES AND RESPONSIBILITIES:
• Develop ultra-fast analytical methods using ultra high-performance liquid chromatography (UHPLC) and high-resolution mass spectrometry (HRMS) for discovery projects to analyze intact proteins, peptides, and other molecules, including small molecules, in complex matrices using state-of-the-art equipment such as Agilent RapidFire/QTOF 6545XT, Sciex UHPLC/ZenoTOF 7600, and Sciex UHPLC/QTRAP 6500+ systems.
• Perform hands-on laboratory research and routine LC/MS analysis of proteins and peptides for identification, quantitation, transformation, conjugation, molecular weight sizing and percent purity in complex matrices.
• Perform hands-on laboratory research and routine LC/MS analysis of small molecules to support animal pharmacokinetic studies for drug discovery projects.
• Generate PK reports using Phoenix 64 WinNonlin software in support of animal PK studies.
• Develop and optimize offline and online sample preparation and cleanup using multiple techniques, such as SPE, Hybrid SPE, PPT, LLE, and SLE, and protein-related digestion and hydrolysis.
• Acquire and process data, interpret results and generate reports using multiple software tools such as Sciex Analyst Software 1.7, Sciex OS 2.0, Agilent MassHunter 10.1, and Agilent Bioconfirm 10.0.
• Troubleshoot and maintain instruments to sustain analytical performance, data reproducibility and reliability to deliver results to project teams within a timely manner.
• Actively and effectively collaborate with Pharmacology team members and cross-functional Discovery teams such as chemistry and molecular biology for experimental design, results interpretation and discussion, creation of hypotheses and conclusions, and planning/recommending next experiments through the interpretation of complex proteins and peptides data.
• Prepare concise and organized analytical protocols and results reports using MS Word and Excel, and effective PowerPoint presentations of experimental results to drug discovery project teams.
• Maintain laboratory electronic notebook (ELN) experimental records current in accordance with company policies and practices.
• Adhere to good health and safety practices and compliance with applicable EH&S safety rules and participate in mandatory safety and other mandatory training programs.

SUPERVISORY RESPONSIBILITIES:
• No supervisory responsibilities

EDUCATION/EXPERIENCE/KNOWLEDGE & SKILLS:
Education:
• Bachelor's degree in related discipline and fifteen years of related experience; or
• Master's degree in related discipline and thirteen years of related experience; or
• PhD degree in analytical chemistry, pharmaceutical sciences, biochemistry, or related discipline and three to six years of related industry experience.

Experience:
• Hands-on experience in protein and peptides analysis by LC/MS/MS and Q-TOF mass spectrometry for the identification and characterization of intact proteins and peptides is required.
• Experience with RapidFire/QTOF 6545XT, Sciex UHPLC/ZenoTOF 7600, and Sciex UHPLC/QTRAP 6500+ systems, is a plus.
• Hands-on experience with LC/MS/MS bioanalytical method development for the quantitation of small molecules in biological matrices is desired.
• Experience using Phoenix 64 WinNonlin software for PK data analysis and report generation.
• Solid experience in sample preparation techniques for protein analysis by LC/MS, and small molecules from animal matrices, such as whole blood, plasma, serum, and tissues.
• Experience with automated liquid handling systems and online sample cleanup by SPE.
• Experience in pharmaceutical drug discovery and development, preparing and analyzing samples, acquiring and processing data, reporting and presenting results concisely and comprehensively within a fast paced and collaborative environment.

Knowledge/Skills:
• Expertise in protein and peptide mass spectrometry.
• Proficiency in Sciex and Agilent LC/MS/MS instruments, ESI and APCI ionization, and QTOF mass spectrometry, as well as related software Sciex Analyst, Sciex OS, Agilent MassHunter, and Agilent Bioconfirm, is essential.
• Knowledge of Phoenix 64 WinNonlin software for PK data analysis and report generation.
• Proficiency with computers including MS Word, MS PowerPoint, and MS Excel is required.
• Demonstrated data analysis skills, problem solving skills and written/verbal communication skills.
• Work effectively, independently, and capable of multi-tasking and prioritizing assignments.
• Excellent collaborative, verbal and written communication skills, data analysis, problem solving, critical thinking, and organization skills
• Detail oriented, can multi-task and work on multiple projects concurrently. Ability to work in a fast-paced organization.
